麦麸低聚木糖产品的分离纯化及定性分析 被引量:3

Separation,Purification and Qualitative Analysis of Xylooligosaccharides Production from Wheat Bran

摘要 对麦麸低聚木糖粗提液进行了活性炭柱层析,采用脱气去离子水及30%乙醇溶液洗脱,洗脱速度0.5mL/min,洗脱曲线峰形较窄而且很对称,可以得到以木二糖~木六糖为主的低聚木糖产品,分离效果较好,因此,活性炭柱层析可以有效地分离纯化麦麸低聚木糖糖液。并且以聚丙烯酰胺凝胶(Bio-Gel P-2)为介质,选用26 mm×800 mm的层析柱,分离活性炭柱层析洗脱浓缩液,对影响分离效果的进样量进行分析后表明,在采用进样量5 mL,洗脱速度0.4 mL/min,脱气去离子水洗脱时,可以分离得到6个明显的单峰曲线,分离效果较好,通过薄层层析图谱可以看出该法能较好的分离纯化出低聚木糖液的各个单一组分。为麦麸低聚木糖产品的分离纯化提供了参考方法,同时也为利用麦麸低聚木糖中各单一组分标样的制备提供了理论依据。 Crude extracts of wheat bran xylooligosaccharides were activated carbon column chromatography,using degassed deionized water and 30% ethanol elution.The elution speed was 0.5 mL/min,and the elution curve of a narrow and very symmetrical peak shape,the xylobiose to xylohexaose could be got primarily xylooligosaccharides products,with better separation.Therefore,the activated carbon column chromatography could be effectively purified wheat bran xylooligosaccharides fluid.And with polyacrylamide gel(Bio-Gel P-2) as the medium,26 mm×800 mm column chromatography was the choice.The elution column chromatography separation of carbon concentrate on the impact of separation of the sample volume for analysis that in using the sample volume 5 mL,the elution speed was 0.4 mL/min.Degassed deionized water elution could isolated 6 distinct single peak curve,with better separation,and through the thin layer chromatogram of the method could be better isolated and purified xylooligosaccharides each single component fluid.Provide the reference method of the separation purification for the wheat bran xylooligosaccharides product,and also for the use of wheat bran xylooligosaccharides in the single component preparation of standard samples to provide a theoretical basis.
