寒热体质大鼠TRPV1通道表达的研究 被引量:11

Study on the expression of TRPV1 in cold and heat constitution rats

摘要 目的:研究寒、热体质大鼠大脑皮质辣椒素受体(TRPV1)通道表达及热痛阈的差异。方法:以两后掌表面皮肤温度为主要指标,结合自主活动及一般情况,从自然群体大鼠中筛选出常体、寒体及热体大鼠,通过光照热痛照射仪测定各组大鼠热痛甩尾潜伏期,比较不同体质大鼠的热痛阈差异。取大鼠大脑皮质,用实时荧光RT-PCR(SYBR GreenⅠ)检测TRPV1通道mRNA表达;蛋白质印迹法(Western Blot)检测TRPV1通道蛋白表达。结果:热体组大鼠热痛甩尾潜伏期时间显著低于常体组和寒体组(P<0.01);热体组大鼠大脑皮质TRPV1通道mRNA相对表达量显著高于常体和寒体组(P<0.01);热体组大鼠大脑皮质TRPV1通道蛋白相对表达量高于寒体组(P<0.05)。结论:寒、热体质大鼠大脑皮质中TRPV1通道mRNA、蛋白表达及热痛阈均存在差异。 Objective:To investigate the differences in expression of TRPV1 and heat pain threshold in cerebral cortex of Cold and Heat constitution rats.Methods:To identify the Normal,Cold and Heat constitution rat from a group of normal rats,the palm skin temperature and the autonomic activities of rat were tested,and the general state of rat were observed also.Heat pain threshold of rats of each group was analyzed by the latency of radiant tail heating.The relative expression of TRPV1 mRNA in cerebral cortex of rats was detected by RT-PCR(SYBR GreenⅠ).The relative expression of TRPV1 protein in cerebral cortex was detected by Western blot.Results:The latencies of radiant tail heating in the rats with Heat constitution are significantly lower than those of Normal or Cold constitution(P0.01).The relative expression of TRPV1 mRNA in cerebral cortex of rats with Heat constitution is significantly higher than those of Normal or Cold constitution(P0.01).The relative expression of TRPV1 protein in cerebral cortex of rats with Heat constitution is significantly higher than those of rats with Cold constitution(P0.05).Conclusion:There are differences in expression of TRPV1 in cerebral cortex and Heat pain threshold of rats with Cold and Heat constitution.
