摘要
目的:研究解毒凉血方对肝卵圆细胞的诱导分化作用及其作用机制。方法:SD大鼠随机分为模型组、解毒凉血方组、正常对照组;通过喂饲2-乙酰氨基芴加2/3肝切除术建立肝卵圆细胞增殖模型;HE染色观察肝组织病理学,透射电镜观察肝卵圆细胞的超微结构,免疫组织化学观察各组大鼠肝组织OV6、ALB、CD34及Notch1的表达和分布。结果:模型组造模过程中至7周末肝组织汇管区周围可见大量的卵圆细胞增生,OV6、CD34、Notch1蛋白与正常对照组比较在肝组织中呈高表达,ALB蛋白较正常对照组表达明显减少;解毒凉血方组肝组织卵圆细胞增生较模型组明显减少,OV6、CD34、Notch1蛋白表达较模型组明显减少(P<0.05)、ALB蛋白表达较模型组明显增加(P<0.05)。结论:解毒凉血方能有效的促进卵圆细胞向成熟肝细胞分化,此作用机制可能与调节Notch信号通路有关。
Objective:To observe the inducement effect and its mechanism of Liangxue Jiedu Fang on hepatic oval cells.Methods:The SD rats were divided into 3 groups:model group,Liangxue Jiedu Fang group and control group.The liver regeneration model was constructed with 2-acet amino fluorene feed plus 2/3 liver resection method.The liver oval cell distribution and expression in every groups were observated by HE stain;the ultrastructural changes were observed by transmission electron microscope.OV6,Notch1,ALB and CD34 distribution and expression in rats liver were observed by immunohistochemical method.Results:Charactered structure in hepatic oval cells was tonofilaments and desmosome juntion.In model group,plenty of liver oval cells proliferation was seen around portal area of hepatic tissue,and OV6,CD34,and the expression of Notch1 protein were higher in liver tissue than those in control group,while ALB protein were lower than those in control group.In Liangxue Jiedu group,hepatic oval cell proliferation was significantly reduced compared with model group,the expression of OV6,CD34,Notch1 proteins were lower than those in model group(P〈0.05),while the expression of ALB protein were higher than those in model group(P〈0.05).Conclusion:Liangxue Jiedu Fang can promote effectively oval cells to hepatic mature cells.Its machanism was related to the regulation of Notch signal pathway.
