血府逐瘀汤对肺血管重构大鼠病理形态学的影响 被引量:8

Pathological morphology study of Xuefu Zhuyu Decoction on pulmonary vascular remodeling in rats

摘要 目的:观察血府逐瘀汤对野百合碱(MCT)诱导肺血管重构大鼠肺血管病理形态学影响,为肺络理论提供实验依据。方法:雄性SD大鼠54只,随机分成正常组、模型组、血府逐瘀汤预防组、血府逐瘀汤治疗组、血府逐瘀汤预防组的硝苯地平对照组和血府逐瘀汤治疗组的硝苯地平对照组,每组9只。采用一次性腹腔注射MCT(60mg/kg)复制肺血管重构模型,血府逐瘀汤预防组和血府逐瘀汤预防组的硝苯地平对照组造模第2天起给药,血府逐瘀汤治疗组和血府逐瘀汤治疗组的硝苯地平对照组造模第15天起给药,直至实验第28天采集组织标本,计算右心肥厚指数(RV/LV+S),右下肺叶组织苏木精-伊红(HE)染色、石蜡切片胶原纤维(Masson)染色观察肺小动脉病理改变。结果:模型组大鼠右心肥厚指数比正常组明显增加(P<0.01),各治疗组右心肥厚指数与模型组比较均明显降低(P<0.05,P<0.01);与正常组比较,模型组肺小动脉管壁变厚、管腔变小,管壁内胶原纤维明显增加(P<0.01),各治疗组与模型组比较肺小动脉管壁变薄,管腔变大,管壁内胶原纤维表达明显减少(P<0.05,P<0.01)。结论:血府逐瘀汤可以明显减轻肺血管重构,其机制可能与调节胶原纤维在肺小动脉的表达有关,研究肺血管重构可以为肺络理论的进一步发展提供部分实验依据。 Objective:The pathological morphology of monocrotaline(MCT) induced pulmonary vascular remolding in rats was observed after treatment with Xuefu Zhuyu Decoction,thus to provide the experimental basis for the illustration of lung-collateral theory.Methods:54 male rats were randomly divided into 6 groups,which were control group,model group,XFZYD prevention group(XFZYD1),XFZYD treatment group(XFZYD2),nifedipine prevention group(N1),and nifedipine treatment goup(N2),9 rats per group.After intraperitoneal injection with MCT(60mg/kg) to produce the pulmonary vascular remolding model,XFZYD and nifedipine were administrated the next day respectively in the XFZYD1 and N1 until the end of the experimental cycle,while rats in the XFZYD2 and N2 were given the corresponding medication from the 15th day of the model construction to the end of the experiment.Rats were sacrificed in the 28 day for tissues and the tissue samples were calculated for the index of right ventricular hypertrophy(RV/LV+S).Small lung arteries were tested for the pathological change by HE staining and Masson staining in the lower right lung lobe.Results:Index of right ventricular hypertrophy in model rats was significantly increased compared with normal control group(P〈0.01),whereas in all treatment groups compared with the model group,indexes were much lower(P〈0.05,P〈0.01).In contrast with the control group,thickening of small pulmonary artery wall,narrowing of lumen,increasing of wall collagen fibers were obviously observed in model group(P〈0.01),but in the treatment groups compared with the model group,wall of small pulmonary arteries were thinning,with the lumen larger,and the wall collagen fibers significantly reduced(P〈0.05,P〈0.01).Conclusion:XFZYT can apparently alleviate the pulmonary vascular remolding,and the possible mechanism is related with regulation of expression of collagen fiber in small pulmonary arteries.Therefore,the research on the pulmonary vascular remolding therefore can provide the basis for further development of lung-collateral theory.
