高效液相色谱法同时测定不同产地、不同炮制时间连翘和连翘叶中5种活性成分的含量 被引量:7

摘要 目的:采用HPLC法测定不同产地、不同炮制时间连翘和连翘叶中芦丁、连翘酯苷A、(+)松脂素-β-D-葡萄糖苷、(+)表松脂素-β-D-葡萄糖苷、连翘苷。方法:色谱柱:Waters Symmetry C18(4.6 mm×250 mm,5μm),柱温25℃,流动相乙腈-0.4%冰醋酸溶液,梯度洗脱,流速1 mL.min-1,检测波长277 nm。结果:芦丁、连翘酯苷A、(+)松脂素-β-D-葡萄糖苷、(+)表松脂素-β-D-葡萄糖苷、连翘苷在检测的范围内线性关系良好,r2均大于0.999 7,方法回收率在97.69%~100.30%之间。结论:此方法简便、准确,快速,为全面评价连翘和连翘叶的质量提供可靠的分析方法。 OBJECTIVE To establish a HPLC method for the simultaneous determination of rutin,forsythoside A,(+)-pinoresinol-β-D-glucoside,(+)-epipinoresinol-β-D-glucoside and phillyrin in different origins and different processing time and the Leaves of Forsythia suspensa.METHODS The chromatographic separation was carried out on a Waters Symmetry C18 column(4.6 mm×250 mm,5 μm) and with gradient elution of acetonitrile and 0.4% aqueous acetic acid at a flow rate of 1 mL·min-1.The detection wave length was 277 nm.RESULTS All the compounds showed good linearity(r20.999 7).The recovery of the assay was in the range of 97.69%-100.30%.CONCLUSION A simple,accurate and credible method was developed and validated for quality contro1 of Forsythia suspense and its' leaves.
