1998～1999年北京地区流感病毒监测及血凝素基因分析

Surveillance and gene analysis of hemagglutinin of influenza virus during the outbreak of 1998-1999 in Beijing area

目的 监测北京地区流感病毒流行 ,探讨其流行规律及血凝素抗原基因变异特征。方法 在 1998年冬北京地区流感病毒暴发流行期间 ,于首都儿科研究所附属医院门诊及北京市防疫站采集标本进行病毒分离、鉴定 ,并对其中 3株病毒的血凝素重链区 (HA1)进行了核苷酸和氨基酸序列分析。结果 共分离到流感病毒 88株 ,经血清学鉴定为甲 3型。与参考株A/Nanchang/ 933/ 95的核苷酸同源性为 (97.0～ 97.7) % ,氨基酸同源性为 (94.2～ 95 .4) % ;与参考株A/Sydney/ 0 5 / 97的核苷酸同源性为 (98.0～ 98.9) % ,氨基酸同源性为 (96 .4～ 97.6 ) %。结论 本次北京地区流行的流感病毒为甲 3型。与近年国内甲 3流行参考株A/Nanchang/ 933/ 95相比 ,其HA1区发生了较大变异 ,而与国际上新出现的A/Sydney/ 0 5 / 97株相近 ,提示本次流行的毒株可能与国外有共同的起源。这是国内首次报道此次流行变异株的核苷酸序列。

Objective Aetiological and molecular epidemiological surveillance of the influenza virus in Beijing area. Methods During the influenza outbreak in Beijing area of 1998-1999, specimens were collected from the Capital Institute of Pediatrics and the Beijing Epidemic Prevention Station. The isolates were identified by using McAb and the HA1 region of 3 isolates were sequenced. Results Eighty eight strains of influenza virus were isolated and identified as subtype A3. The analysis of the nucleotides and deduced amino acids suggested that the prevalence strains had experienced some variations as compared with the national reference strain A/Nanchang/933/95 of recent years, and showed high similarity with the new international prevalence strain A/Sydney/05/97. Conclusions The prevalence strains of the influenza outbreak of 1998-1999 in Beijing area were subtype A3 and there is certain antigenic drift in influenza virus in Beijing area. The results suggest that they may be Sydney-like strains and has the same origin with that new international prevalence strain.
