摘要
目的 探讨人工合成的 Cp G ODN作为佐剂对乙肝病毒基因疫苗诱导小鼠产生抗 HBs的影响 .方法 构建编码乙型肝炎病毒 (HBV)表面抗原蛋白 S的重组真核表达质粒p CR3.1- S作为 HBV基因疫苗 ,人工合成含 Cp G motif的硫代磷酸寡核苷酸作为佐剂 ,以 BAL B/ c小鼠作为实验动物进行免疫接种 ,采用 EL ISA法检测免疫小鼠的抗 HBs应答 .结果 与生理盐水作为佐剂组相比较 ,应用 Cp G ODN组小鼠产生更强的抗 HBs反应 (P<0 .0 5 ) .结论 Cp G ODN作为佐剂可促进 HBV基因疫苗诱导 BAL B/ c小鼠产生抗 HBs应答 .
AIM To investigate the effects of CpG ODN as adjuvants on the production of anti HBs in mice immunized with hepatitis B virus (HBV) gene vaccines. METHODS Recombinant eukaryotic expression plasmids of pCR3.1 S encoding S proteins of HBV were constructed as gene vaccines. Phosphorothioate oligodeoxynucletides containing one CpG motif were sythesized as adjuvants. Then BALB/c mice were inoculated so that the immune responses to HBV S proteins could be studied. Anti HBs antibodies were detected by ELISA assays. RESULTS Immunized mice with CpG ODN as experiment adjuvants showed higher anti HBs than ones with saline as control adjuvants ( P <0.05). CONCLUSION CpG ODN can act as adjuvants to increase the production of anti HBs induced by HBV gene vaccines.
