摘要
目的 观察新的二氢吡啶类钙拮抗剂拉西地平对大鼠肾性高血压的治疗作用 .方法 采用一肾一包扎型肾性高血压模型 ,测定不同条件下动物血压、心脏重量、肾血流量(RBF)和肾小球滤过率 (GFR) .结果 术后 4wk开始给予拉西地平 (2 0 mg· kg- 1 ,ip,b.i.d) ,可显著降低心脏湿重与体质量比 (HWW/ BW)和左心湿重与体质量比 (L VWW/ BW)(P<0 .0 1) ,改善心肌肥厚症状 .静脉连续输入拉西地平和Nifedipine,在 (1~ 10 ) μg·kg- 1·min- 1范围内 ,产生显著的降压作用 ,并表现出剂量依赖性 ,拉西地平降压效应明显大于等剂量 Nif.同时发现 ,拉西地平和 Nif在 1μg· kg- 1·min- 1时 ,均可显著增加 RBF和 GFR(P<0 .0 5 ) ,且拉西地平增加 RBF的程度比 Nif显著 .但这一效应在 3,10μg·kg- 1 · min- 1 时 ,由于血压过低而逆转 .
AIM To study the therapeutic effects of a novel dihydropyridine calcium antagonist, lacidipine, on renal hypertension of rats. METHODS Using one kidney one clip renal hypertensive model, we determined the arterial pres-sure, heart weight, renal blood flow (RBF) and glomerular filtration rate (GFR) at varied treatment. RESULTS Given from 4 weeks after operation, lacidipine (20 mg·kg -1 , ip, b.i.d) significantly decreased the ratios of heart wet weight to body weight (HWW/BW) and left heart wet weight to body weight (LVWW/BW) ( P <0.01), reversing myocardial hypertrophy. At the dose of 1~10 μg·kg -1 ·min -1 lacidipine and Nifedipine induced hypotensive reaction in the dose-dependent manner, and lacidipine showed more potential hypotensive reaction than that of Nifedipine at the same dose. In the meantime lacidipine and Nifedipine notably increased RBF and GFR at 1 μg·kg -1 ·min -1 ( P <0.05) , while lacidipine showing stronger than Nifedipine. However, this effect was reversed at 3 and 10 μg·kg -1 ·min -1 for the excessive hypotension. CONCLUSION Lacidipine exerts satisfying therapeutic effects on renal hypertension of rats.
