摘要 目的 确立血脾屏障的组成及形态构筑 ,探讨血脾屏障在脾功能中的作用 .方法 采用健康昆明株小白鼠 10 0只 ,随机分成 10组经尾静脉注射 5 %墨汁混悬液 (0 .2~ 0 .3m L /只 ) ,分别在 1,3,6 ,12 h及 1,3,5 ,7,14,2 8d取脾进行组织学、免疫组化第 因子相关抗原染色 ,在光镜、电镜下观察 ,同时检测内皮细胞、巨噬细胞吞噬墨汁颗粒的数量 .结果 脾血窦内皮细胞、巨噬细胞对第 因子相关抗原呈阳性反应 .注射墨汁混悬液后 3h,6 d内皮细胞、巨噬细胞内可见墨汁颗粒 ,在 2 4h,3d组最多 ,随后逐渐减少 ,墨汁颗粒主要集中在红髓及边缘区 .结论 血脾屏障由脾窦内皮细胞、基底膜、巨噬细胞组成 。 AIM To study the structure and morphology of blood-spleen barriers and to explore their roles in spleen function. METHODS One hundred healthy Kunming mice were randomized into ten groups. Each mouse was injected with chinese ink (0.2~0.3 mL, 5%) from the tail vein. Histological and immunohistochemical staining techniqueswere used to inspect the barriers at different time points after the injection. Pieces were observed under light and electron-microscopes. The amount of the pinocytotic and phagocytic ink particulates in the endothelial cells and macrophages was measured under the light microscope. RESULTS Splenic sinus endothelial cells and macrophages were factor Ⅷ-related-antigen positive. The ink particulates could be found in endothelial cells and marcrophages three and six hours after injection. In 24 h and 3 d groups there were more ink particulates than in any other group. These cells were localized in red pulp and marginal zone. CONCLUSION Blood-spleen barrier is composed of splenic sinus endothelial cells, basement-membrane and macrophages. They are located in red pulp and marginal zone.
