Effects of 6-BA and NAA in Different Concentration Proportions on PLB Induction of Cymbidium Hybridum

摘要 以大花蕙兰茎尖为外植体,研究6-BA与NAA浓度梯度配比对大花蕙兰原球茎的诱导、增殖及分化的影响.结果表明:6-BA 1.0mg/L+NAA 0.5mg/L浓度配比下,大花蕙兰茎尖存活率达到93.3%;6-BA0.8mg/L+NAA 0.4mg/L浓度配比下,原球茎诱导率达到85.8%;6-BA 1.0mg/L+NAA 0.3mg/L浓度配比下,原球茎增殖系数达到4.59;6-BA 0.8mg/L+NAA 0.5mg/L浓度配比下,原球茎出芽率达到87.5%.首次报道采用6-BA 0.8mg/L+NAA 0.5mg/L浓度配比组合适合大花蕙兰茎尖培养,可同时获得较高的原球茎诱导率(80.8%)和出芽率(87.5%),比目前已报道的最佳原球茎诱导率(72.7%)和出芽率(76.7%)提高8%以上. Shoot-tips from Cymbidium hybridum were used as the explants to investigate the effects of 6-BA and NAA in different concentration proportions on PLB (protocorm-like body) induction, propagation and differentiation. The results show that the survival rate of shoot-tips is 93.3% in the concentration proportion of 1.0 mg/L6 BA + 0.5 mg/L NAA, the induction rate of PLB is 85.8% in the concentration proportion of 0.8 mg/L 6-BA + 0.4 mg/L NAA, the propagation rate of PLB is 4.59 in the concentration proportion of 1.0 rng/L 6-BA + 0.3 mg/L NAA, and the budding rate of PLB is 87.5 % in the concentration proportion of 0.8 mg/L 6-BA + 0. 5 mg/L NAA. The optimized concentration proportion of 0.8 mg/L 6-BA + 0.5 mg/L NAA can result in 8% higher ratio of PLB induction (80.8o//oo) and differentiation (87. 5‰) than that with currently reported 72.7% and 76.7% respectively.
