摘要
分别采用传统组织块法、传统酶消化法和优化组织块法进行新生SD大鼠原代成骨细胞体外分离和培养,并对培养出的细胞予以鉴定和比较。优化组织块法可短期内培养出大量纯度高的成骨细胞,具有典型的形态特征和成骨能力,碱性磷酸酶(ALP)染色、Ⅰ型胶原免疫荧光均呈阳性,ALP检测活性可达92%。该法优于其他两种方法,提供了一种切实可行、操作简单的体外原代成骨细胞培养方法。
The original generation of osteoblasts of 24-hour old SD rats were isolated and cultured in vitro with three methods--the traditional tissue plant method, traditional enzyme digestion method, and modified enzymatic digestion method. The ostcoblasts generated by 3 methods were identified and compared with each other at last. The modified enzymatic digestion method can train a lot of high purity.osteoblasts in a short term, which cells had typical morphological and biological characteristics of osteoblasts. The ALP dyes and type I collagen immunity fluorescence were all positive. The activity of ALP reached 92%. This method is better than the other two, and provides a feasible, simple operation of the generation osteoblast training method in vitro.
