摘要
目的检测以多功能心腔内导管对活体犬行心肌内注射的可行性和安全性。方法将12只健康杂种犬随机分为3组,在超声心动图模式下引导多功能心腔内导管进入左心室,在心腔内超声模式下监控注射针插入左心室前壁及室间隔,并注射伊文思蓝染料,剂量分别为0.4ml、0.8ml和1.2ml。3h后处死动物取出心脏,观察注射点与超声图像的位置关系,计算染色心肌的体积,并对各组进行比较。结果多功能心腔内导管顺利进入心室,能对心肌结构进行识别,有效监控注射针对心肌进行靶向注射。各组染色心肌体积差异有统计学意义。注射相同剂量的染料在左心室前壁与室间隔形成的染色心肌体积差异无统计学意义。结论本研究设计的多功能心腔内导管系统与超声心动图整合后基本达到预期目标,可能为心肌内干细胞或基因注射提供更为安全、有效的途径。
Objective To investigate the feasibility and safety of transendocardial Evan's blue injection in canine using a self-developed multifunctional intracardiac catheter. Methods Twelve mongrel canines were randomly divided into three groups. Under the mode of echocardiography, the catheter was delivered into the left ventricle. Under the intracardiac ul- trasound mode, 0.4 ml, 0. 8 ml or 1.2 ml Evan's blue were sequentially injected into the left ventricular anterior wall and the interventricular septum. Electrocardiographic recordings were obtained during the whole procedure. Three hours after injection, animals were sacrificed and the hearts were harvested. The size of stained myocardial was evaluated. Results Under the guidance of echocardiography, the catheter was inserted to the ventricle. The catheter provided the structural in- formation and guided proper needle position within the target myoeardium. There were significant differences in the size of stained myocardial produced by injection of different amounts of Evan's blue infusion, but no significant differences in the size of stained myocardial by injection of the same amounts of Evan's blue infusion was found in the left ventricular anterior wall and the interventricular septum. Conclusion These multifunctional intracardiac catheter system integrated with echo- cardiography meets the requirements of primary design, may provide a safe and effective way for transendocardial injection in future.
