摘要
对硫酸妥布霉素生产中发酵液的水解、离子交换层离、成盐等条件进行了研究。结果表明 :6" -O-氨甲酰妥布霉素水解形成妥布霉素的反应为一级反应 ,其水解介质最佳 p H值为 1 0。用氨水作为离子交换层离妥布霉素解吸剂的最佳浓度为 0 .1 mol/ L。妥布霉素碱转盐应采用高限量计算的硫酸。按上述条件生产的硫酸妥布霉素产品附合 USP标准。
Among the procedure of tobramycin production, hydrolysis of the fermantation solution, purification by ion exchange chromatography and salt formation has been studied. The results show that the hydrolysis of 6' O carbamyl tobramycin to form tobramycin agree with the first order reaction; the best pH of hydrolysis medium should be 10. The proper concentration of ammonia aqua which used as eluant in the ion exchange chromatography is 0.1 mol/L. The quantity of sulfuric acid used to transform tobramycin base to tobramycin sulfate should be calculate according to the high limit. The tobramycin sulfate produced according to the above condition, will conform with USP standard.
