高效液相色谱“柱切换法”测定血液及腹腔液中5-氟尿嘧啶的浓度 被引量:1

摘要 目的 :评价高效液相色谱柱切换法在测定体液中 5 -氟尿嘧啶 (5FU)浓度的优点。方法 :高效液相色谱柱切换法测定血及腹腔液中 5FU浓度。结果 :标准曲线的回归方程为Y =9 6 9× 10 -6X - 1 33 ,r =0 9998,P <0 0 0 0 1;体液测定 5FU浓度的RSD为 0 2 %~ 5 6 % ,但是浓度小于或等于 0 0 5 μg·mL-1时 ,RSD为15 7%。体液中 5FU的回收率为 88 87%~ 116 8%。测定的最小浓度为 0 0 5 μg·mL-1。结论 :高效液相色谱柱切换法有样品处理简捷快速 ,灵敏度高 ,重复性好等优点 ;在临床药物监测时 。 Objective:To evaluate the advantages of HPLC Column Replacement Technique in the determination of 5FU concentrations in biological fluids.Methods:Blood and peritoneal fluid concentration of 5FU were determined by HPLC Column Replacement Technique.Results:The calibration curve was linear in the range of 0 05~500 μg·mL -1 The standard curve equation was Y =9 69×10 -6 X -1 33, r =0 999 8, P < 0 000 1 .Reproducible blood and ascites concentrations were determined with RSD ranging from 0 2% to 5 6% RSD was 15 7% for concentrations equal to 0 05 μg·mL -1 The recovery rate ranged from 88 87% to 116 8% The minimal detectable concentration was 50 μg·L -1 Conclusions:The HPLC Column Replacement Technique has many advantages:(1)extraction procedure is easier and less time consuming;(2)sensitivity is high;(3)reproducibility is good.HPLC Column Replacement Technique can have good application in the monitoring of therapeutics drugs.
