摘要
目的:对1株静置培养产生凝胶样物质的菌株CIs51进行鉴定,并对其产物进行分析,确定其工业化生产前景。方法:以CIs51为目的菌株,根据其形态学特征、生理生化特性、16SrDNA序列分析,鉴定其种属。对CIs51所产凝胶干膜进行纤维素酶水解试验、红外光谱分析、X-射线衍射分析,确定其主要成分及性质。研究菌株在柑橘果渣水培养基中的凝胶干膜产量、传代稳定性等,确定该菌株的工业化生产前景。结果:根据形态观察、生理生化特性与16SrDNA序列分析结果,CIs51被鉴定为汉逊氏葡糖酸醋杆菌(Gluconac etobacter hasenii),命名为汉逊氏葡糖酸醋杆菌CIs51。该菌株所产凝胶膜中纤维素含量达94%;红外光谱分析表明其主要成分为β构型葡聚糖,为I型纤维素,结晶度为75%。CIs51在柑橘果渣水培养基中生长良好,纤维素产量达10.17g/L,传代稳定性好。结论:汉逊氏葡糖酸醋杆菌CIs51为新型细菌纤维素高产菌株,适宜在柑橘果渣水培养基中生产细菌纤维素,具有工业化生产的潜力。
Objective: To identify a gel-like membrane producing strain CIs51, investigate characteristics of the gel- like substance and provide feasible technology for industrial production. Methods: CIs51 was identified through morphology observation, physiological and biochemical characteristics and 16S rDNA sequence analysis. The gel-like membrane pro- duced by CIsS1 was analyzed through cellulase hydrolysis experiments, infrared (IR) spectrum and X-ray diffraction (XRD) analysis to determine its main components and characteristics. The yield of gel-like dry sheet and passage stabil- ity in citrus dregs solution media were also investigated to determine its industrial production prospect. Results: CIs51 was identified as Gluconacetobacter hasenii CIs51 through morphology observation, physiological and biochemical charac- teristics as well as results of 16S rDNA analysis. Cellulose accounted for a proportion of 94% in the gel membrane. Re- suits of IR spectrum and XRD indicated that it was β structure glucoscan and I type bacterial cellulose, crystallinity in- dex was 75%. CIs51 grew well and maintained passage stability in citrus dregs solution media with the yield of cellulose reaching to 10.17 g/L. Conclusions: Gluconacetobacter hasenii CIs51 was a new isolate with high yield of cellulose, which grew well in citrus dregs solution media and had potentiality of industrial production.
