松江鲈MyoD基因cDNA部分片段的序列和表达特征分析

摘要 生肌决定因子MyoD是生肌调节因子MRFs家族的一个重要成员,在基因转录调控中起着重要的作用.本实验通过RT-PCR和3’-RACE等方法,以松江鲈肌肉总RNA为模板,扩增出957bp的MyoD基因部分序列,包含部分编码区长567bp,编码翻译188个氨基酸残基,3’-UTR区长390bp.系统发育分析表明,松江鲈MyoD与奥尼罗非鱼亲缘关系最近.不同组织半定量分析显示,松江鲈MyoD在各种组织中均有表达,在心脏中表达量最高,预示着在不同组织中该基因的功能广泛. Myogenic determination gene is an important member of the myogenic regulation factors (MRF) family, which plays an important role in the specific gene transcription. A 957 bp long MyoD cDNA sequence in Trachidermusfasciatus was obtained by using RT-PCR and 3'- RACE method, which included a 390 bp 3'-UTR, and partial ORF 567bp encoding 188 amino acid peptide. MyoG gene expression was detected in all six examined tissues, and highly expressed in heart. It suggested that the MyoD gene might contribute to a wide range of biological functions.
