摘要
[目的]研究粗叶悬钩子(rubus alceaefolius poir,RAP)中生物碱类化合物在大鼠各肠段的吸收动力学特征。[方法]采用大鼠在体肠吸收模型,通过紫外分光光度法测定粗叶悬钩子总生物碱的含量及高效液相色谱法比较灌流前后化学成分差异,研究大鼠各肠段的吸收特性。[结果]粗叶悬钩子总生物碱在十二指肠、空肠、回肠和结肠中药物的吸收百分率(%)分别为(3.02±0.25)、(3.42±0.62)、(3.22±0.91)和(2.91±0.92)。[结论]粗叶悬钩子在小肠上段吸收良好,总生物碱中多种成分在肠段可能发生代谢转化。
[Objective]To investigate the absorption characteristics and mechanism of the total alkaloids of R. alceaefolius Poir for various intestinal segments. [Methods]The intestine in rats was cannulated for in situ recirculation. The total alkaloid con- tent was measured by Ultraviolet spectrophotometry. The chemical composition differences were compared before and after perfu- sion by high performance liquid chromatography(HPLC) to study the intestinal absorption characteristics. [Results]The total al- kaloids of R. alceaefolius was metabolized quickly in the four intestinal segments. The permeability coefficients were 3.02± 0.25, 3.42±0.62, 3.22±0.91 and 2,91±0.92 at duodenum, jejunum, ileum and colon respectively. [Conclusion] Some compositions of the total alkaloids ofR. alceaefolius show metabolic transformation.
