脂质过氧化内皮细胞cAMP水平与单核细胞粘附关系的研究 被引量:1

摘要 目的 :研究内皮细胞脂质过氧化损伤后环磷腺苷酸 (c AMP)含量变化及其与单核细胞粘附的关系。方法 :用氧化剂联胺作用于培养人脐静脉内皮细胞 ,诱发脂质过氧化损伤后 ,以生化方法检测内皮细胞 c AMP含量变化 ,光镜观察计数单核细胞对内皮的粘附 ,并用抑制 c AMP因素 Galanin观察其对单核细胞粘附的影响。结果 :适当浓度的联胺可引起内皮脂质过氧化损伤。脂质过氧化损伤后内皮的 c AMP含量显著增加 (P <0 .0 5 ) ,单核细胞粘附显著增加 (P <0 .0 1)。Galanin使单核细胞粘附下降。结论 :脂质过氧化导致的单核细胞对内皮粘附的增加可能是以 c AMP作为第二信使实现的。 Objective: In order to investigate the association between cyclic adenosine monophosphate (cAMP) content of endothelial cell (EC) and the adhesion of monocyte (MC) to EC, and to further understand the mechanism of messages transduction. Methods: cAMP content of EC was measured by radioimmunological assay after the addition of diamide. The number of MC adhesion was accounted under light microtelescope. In another series of experiments, cAMP inhibiting agent Galanin was added, then cAMP and MC adhesion were measured. Results: Diamide could lead to the injury to EC as a result of lipid peroxidation. cAMP content of EC increased after diamide was added. MC adhesion to EC was paralleled to the changes of cAMP content. Conclusion: cAMP as a second messenger might play an important role in the mechanism of increasing adhesion of MC to EC.
