摘要
目的:实验观察雌激素对软骨内成骨过程的影响,初步探索雌激素对骨作用的分子机制。方法:采用体外培养小鼠胚胎长骨模型,通过免疫组织化学和分子原位杂交技术,测定雌酚酮干预后长骨骺板静止区、增殖区和肥大区c-fos mRNA、c-jun mRNA及其表达产物的变化。结果:培养介质中加入0.1 μmol/L雌酚酮培养48h后,长骨骺板静止区、增殖区和肥大区c-fos mRNA、c-jun mRNA及其表达产物蛋白均不同程度上调。结论:雌激素可能通过影响软骨细胞对c-fos和c-jun的表达而影响启动序列中含有AP-1结合位点的基因如TGF-β、IGF-Ⅰ、Ⅱ等的表达,从而影响软骨内成骨过程。
Objective:To study the effects of estrogen on the endochondral bone formation and to explore the mechanism by which estrogen influence on the bone. Methods:The long bones of fetal mice of 16 days were cultured in BGJ medium in a continu-ously rotating device and treated with 0.1 μmol/L estrone. Every 24 hours,a mixture of 45% O2 ,5% CO2 and 50% N2 in culture bottles was changed. After 48 hours' culture, the c-fos mRNA and c-jun mRNA in growth plate were determined by in situ hybridization. The immunohistochemical analysis of c-fos protein and c-jun protein was also performed. Results:The levels of c-fos mRNA and c-jun mRNA and their expressing product proteins in resting zone, proliferative zone and hypertro-phic zone were all upregulated. Conclusion: The results suggested the possibility that estrone enhanced endochondral bone formation by upregulating the expression of c-fos and c-jun in chondrocyte,and their expressing proteins in turn rapidly reg-ulated expression of other genes related to bone growth which were at AP-1 site,such as TGF-β and IGF-I, II.
