摘要
目的应用SD乳鼠器官型海马脑片探讨羟基红花黄色素A(HSYA)对正常及缺血损伤后细胞再生的影响。方法制备P3 SD乳鼠器官型海马脑片,随机分为模型组和正常对照组,每组包含3个亚组,造模前及造模后1d、3 d和6d分别通过倒置显微镜及免疫荧光染色观察脑片生长情况,计数新生细胞并进行各组间的比较。结果模型组新生细胞数在造模后的各个时间点均显著少于对照组(均P<0.01)。HSYA干预后,两组新生细胞均有显著性增加(均P<0.01)。大剂量组新生细胞的增殖显著高于小剂量组(均P<0.01)。新生细胞(DAPI+,BrdU+和Nestin+)多分布于海马,DG区最明显,排列呈条索状,向皮层迁移。结论 HSYA在正常及缺血情况下均可以促进SD乳鼠器官型海马脑片细胞再生。
Objective To investigate the effects of hydroxysafflor yellow A (HSYA) on nerve regeneration of the organotypic hippocampal slices from neonatal SD rats. Methods The organotypie hippocampal slices (300p.m) from postnatal 3rd day (P3) SD rats were prepared and randomly divided into two groups :model group(MG) and control group(CG). Each group was cultured in the same condition and received the same treatment. After that, new cells were observed under invert microscope or detected by immuno fluorescence staining at different time points. Counted new cells and made the comparison between groups. Results At each time point after modeling,new cells in MG were significantly less than CG ( all P 〈0.01 ). After HSYA intervention ,new cells were significantly increased in all groups (all P 〈 0.01 ). And new cells of high - dose group were significantly more than small - dose groups ( allP 〈 0.01 ). New cells ( DAPI + , BrdU + and Nestin + ) were distributed in the hippocampus,and were more obvious in DG, arranged in cords, migrate to the cortex. Conclusion HSYA can contribute to nerve regeneration of organotypic hippocampal slice from neonatal SD rats in normal or ischemic condition.
