摘要 目的建立重楼植物愈伤组织培养的快速繁殖体系。方法文章以黑籽重楼不同处理的芽为外植体,研究了不同消毒时间和激素配比对其外植体愈伤组织诱导与增殖的影响。结果去芽鞘的芽外植体用升汞消毒8 min时,其污染率较低为(11.97±0.91)%、而启动率却较高为(84.82±2.59)%;诱导去芽鞘的芽外植体产生愈伤组织的最佳培养基配方为MS+6-BA 1.0 mg/L+IAA2.0 mg/L,其愈伤组织增殖的最佳培养基配方为MS+6-BA0.5 mg/L+NAA 1.0 mg/L。结论实验实现了黑籽重楼愈伤组织的诱导和快速增殖,并为进一步研究重楼属其它植物的生物技术研究奠定了技术基础。 Objective To establish the best tissue culture system of Paris thibetica for fast propagation. Methods The bud of Paris thibetiea's was used as explants. The effect of different plant hormones and length of disinfection time were analyzed. The result was evaluated by the callus induction rate and propagation rate. Results The contamination rate was only (11.97 ± 0.91 ) % , activation rate was (84.82 ± 2.59) % upon the mercury chloride treatment for 8 minutes. The MS medium supplied with 1.0 mg . L - 1 6 - BA and 2.0 mg . L -1 IAA gave the highest induction rate. The MS medium supplied with 0.5 mg/L 6 - BA and 1.0 mg/L , NAA showed the highest propagation rate. Conclusion The callus induction and rapid proliferation of Paris thibetica was obtained in this study ,which has laid the technical foundation for further biotechnology study on other Paris plants on hiotechnology research.
