摘要
目的:探讨孕妇妊娠期糖尿病患者胎盘中HGF及其受体和RAGE的表达及意义。方法:对各项临床资料完整的晚孕GDM孕妇53例(其中由外院转入孕期未行规律治疗者或晚孕检查发现血糖异常诊断为GDM的23例)为观察组,选择同期来产检并分娩的正常晚孕孕妇30例为对照组。采用免疫组织化学方法测定两组胎盘中HGF、c-Met和RAGE 3种蛋白表达位点及表达水平,采用逆转录-聚合酶链反应(RT-PCR)测定三者的mRNA水平。结果:凝胶图像分析显示,GDM组HGFmRNA、c-Met mRNA和RAGE mRNA相对含量分别为0.232±0.065、0.172±0.063及0.673±0.134,与对照组相比差异有统计学意义(P<0.01);三者在观察组胎盘中的表达量分别为8.017±1.327、9.417±1.360及12.398±2.381,与对照组相比差异有统计学意义(P<0.01)。结论:妊娠期糖尿病患者胎盘中HGF-mRNA及其受体c-Met mRNA的水平及表达水平显著低于对照组,而RAGE mRNA及表达水平高于对照组(P<0.01),这对妊娠期糖尿病的诊断预防及治疗均具有重要意义。
Objective: To explore the expressions and significances of human growth factor (HGF) and its receptor (c - Met), RAGE in placenta of patients with gestational diabetes mellitus (GDM) . Methods: A total of 53 GDM patients during the third trimester of pregnancy with intact clinical data were selected as observation group, including 23 GDM patients who were transferred from other hospitals and had no regular treatment or who were diagnosed as GDM by finding blood glucose abnormality during the third trimester of pregnancy ; 30 normal pregnant women during the third trimester of pregnancy who went to the hospital for prenatal examination during the same period were selected as control group. Immcnohistochemical method was used to detect the expression sites and expression levels of HGF, c -Met, and RAGE in placental tissues of the two groups ; the levels of HGF mRNA, c - Met mRNA, and RAGE mRNA in the two groups were detected by RT - PCR. Results: Gel image analysis showed that the relative contents of HGF mRNA, c - Met mRNA, and RAGE mRNA in observation group were (0.232 ±0.065), (0. 172 ±0.063), and (0. 673 ±0. 134), respectively; compared with control group, there was statistically significant difference ( P 〈 0. 01 ) . The expression levels of HGF, c - Met, and RAGE in placenta of observation group were (8. 017 ± 1. 327), (9. 417 ± 1. 360), and ( 12. 398 ±2. 381 ) , respectively; compared with control group, there was statistically significant difference (P 〈 0. 01 ) . Conclusion: The relative contents of HGF mRNA and c - Met mRNA, the expression levels of HGF and c - Met in placenta of patients with GDM were statistically significantly lower than those in control group, while the relative content of RAGE mRNA and expression level of RAGE in placenta of patients with GDM were statistically significantly higher than those in control group, which had im- portant significances for diagnosis, prevention, and treatment of GDM.
