3种典型海洋微藻的细胞增殖特性研究 被引量:3

摘要 目的研究硅藻门的三角褐指藻Phaeodactylum tricornutum、甲藻门的东海原甲藻Prorocentrum donghaiense和金藻门的球形棕囊藻Phaeocystis globosa的细胞增殖情况,为认识其基本生物学特性,推动海洋微藻在医药保健食品行业的应用提供参考。方法选取上述3种海洋微藻为生物材料,以添加f/2营养液的人工海水为培养介质,在植物培养箱中利用三角瓶开展为期2周的一次性培养实验。利用倒置显微镜和血球计数板观测微藻细胞密度,采用最小二乘法拟合法计算微藻日生长速率。结果 3种海洋微藻的细胞生长均经历缓慢期、快速期和平稳期3个阶段;实验结束时,三角褐指藻、东海原甲藻和球形棕囊藻的细胞密度分别约为1.126×107、1.167×107和1.210×107个/mL;其最大生长速率分别出现在第2天(1.11/d)、第4天(1.13/d)和第3天(1.02/d)。结论 3种海洋微藻的细胞增殖具有多样性;它们的最大细胞密度和日生长速率出现的时间或程度均存在一定的差异,提示在进行经济微藻培养生长过程中必须考虑各藻种的细胞增殖特性。 Objective To investigate the cell proliferation characteristics of three marine microalgae Phaeodactylum tricornutum,Prorocentrum donghaiense and Phaeocystis globosa,hence to understand their basic biological feature,which is of great importance in promoting the development of marine microalgae in the field of health care.Methods P.tricornutum,P.donghaiense and P.globosa were cultured in a plant growth chamber in sea water enriched with f/2 medium for 2 weeks.Cell density was monitored using a hemacytometer under microscopy,and daily growth rate was obtained by least square method.Results The three microalgae showed the slow growth stage,rapid growth stage and steady growth stage.In the end of the experiments,the cell densities were 1.126×10^7,1.167×10^7 and 1.210×10^7 cells/mL for P.tricornutum,P.donghaiense and P.globosa,while their maximal growth rate occurred on the second day(1.11/d),fourth day(1.13/d) and third day(1.02/d).Conclusion Cell proliferation characteristics are different for the three marine microalgae.The diversities in timing and extent of maximal cell density and growth rate should be taken into account when marine microalgae are cultured for study or application.
