乳胶凝集试验检测鸡传染性鼻炎血清抗体的研究

Study of Latex Agglutination Test for the Detection of Serum Antibodies against Infectious Coryza in Chickens

将 A型和 C型鸡副嗜血杆菌培养后 ,菌落计数 ,离心计数 ,离心后用 0 .0 1M p H7.4磷酸盐缓冲液洗涤菌体 2次。经过对致敏温度、致敏时间、致敏乳胶的菌体浓度及菌体的处理方式等条件的选择 ,建立了检测鸡传染性鼻炎血清抗体的乳胶凝集试验。与琼脂扩散试验相比 ,特异性一致 ,但更加敏感 ,且操作简便、快速。结果说明 ,该方法有较好的应用前景 。

Haemoplius paragallinarum (HPG) serotype A and serotype C were cultured,the colonies of bacteria were counted,the bacterial suspension was centrifuged and supernatant was decanted.The precipitate was washed two times with 0.01M pH7.4 PBS.After determining the sensitization conditions such as temperature,time,bacterium concentration and ways of treating bacterium in the course of preparation of the antigen,a latex agglutination test (LAT) for detecting serum antibodies against infectious corzya(IC) was developed.Compared with agar gel diffusion assay, the LAT,with simplicity,shared the same specificity but more sensitive.The result showed that the latex agglutination test had wide application in clinical practice,especially in the rapid diagnosis of infectious corzya (IC) in the field.
