摘要
目的探讨转录因子T-bet,GATA-3和Foxp3在玫瑰糠疹发病中的作用机制。方法采用SYBR GreenI实时荧光定量逆转录-多聚酶链反应技术检测20例玫瑰糠疹患者和22例正常人外周血单个核细胞中T-bet,GATA-3及Foxp3 mRNA的表达水平。结果玫瑰糠疹患者T-bet mRNA RQ值为(0.83±0.57),其表达水平高于正常对照组(0.52±0.36),两者比较差异有统计学意义(P<0.05)。玫瑰糠疹患者GATA-3及Foxp3 mRNA RQ值分别为(0.68±0.29),(0.92±0.61),其表达水平低于正常对照组的(1.22±1.02),(1.48±0.93),差异均有统计学意义(P均<0.05)。结论玫瑰糠疹患者存在T-bet/GATA-3升高,Foxp3降低,Th1细胞亚群优势表达可能性很大,细胞免疫异常可能在玫瑰糠疹发病机制中起着重要作用。
Objective To explore the roles of transcription factors T-bet , GATA-3 and Foxp3 in pathogenesis of pityria- sis rosea (PR). Methods SYBR Green 1 Real-time fluorescence quantitative RT-PCR was employed to de tect the expression of T-bet, GATA-3 and Foxp3 mRNA in peripheral Blood Mononuclear cells from 20 patients with PR and 22 healthy controls. Results The gene T-bet mRNA RQ values in PR group and the control group were (0.83±0.57) and (0.52 ±0.36), and the expression'level of T-bet mRNA in the PR group was higher than that in the control group ( P 〈 0.05 ). The gene GATA-3 mRNA RQ values in PR group and the control group were (0.68 ± 0.29) and ( 1. 22 ±1.02), and the expression level of GATA-3 mRNA in the PR group was lower than that in the control group(P 〈0.05). The gene Foxp3 mRNA RQ val ues in PR group and the control group were (0.92 ± 0. 61 ) and (1.48 ±0. 93 ), and the expression level of Foxp3 mRNA in the PR group was lower than that in the control group(P 〈 0.05). Conclusion The ratio of T-bet/GATA-3 increases in patients with PR, while Foxp3 decreases. There is a high possibility that Thl cells subsets are predominant and the abnormality of cellular immunity may play an important role in the pathogenesis of PR.
