摘要
目的探讨Galectin-1在大肠癌细胞系中的表达情况,以及与大肠癌侵袭、转移的关系。方法用Real time-RT PCR法和免疫组织化学法分析Galectin-1mRNA及蛋白在5种大肠癌细胞系的表达情况;用Boyden小室法比较5种大肠癌细胞系的侵袭转移能力。结果 Galectin-1在大肠癌转移灶来源的细胞系中的表达高于大肠癌原发灶来源的细胞系(P<0.01);Boyden小室的结果显示大肠癌转移灶来源的细胞系的侵袭,转移能力高于大肠癌原发灶来源的细胞系。结论 Galectin-1的高表达可能与大肠癌的侵袭转移有一定关系。
Objective To explore the expression of Galectin-1 gene in colorectal cancer cell lines and its correlation with tumor metastasis.Methods Expression of Galectin-1 in colorectal cancer cell lines was detected by Real time-RT PCR and imunohistochemistry.In vitro invasiveness was determined by Boyden chamber assay.The correlation of Galectin-1 expression with the invasive ability was also analyzed.Results Galectin-1 was highly expressed in LoVo and SW620.In vitro,cell invasivion demonstrated that LoVo and SW620 had a higher invasive ability than LS174T,SW480 or HT29.Conclusion Galectin-1 may play an important role in invasion and metastasis of colorectal cancer.
