摘要
用不同浓度的低聚木糖处理大豆,研究低聚木糖对大豆生长和花叶病毒病(SMV)抗性的影响,温室实验结果表明:低聚木糖溶液处理大豆种子18 d后与对照相比,50 mg.L-1低聚木糖对大豆幼苗的株高、鲜重和主根长有明显的促进作用。低聚木糖喷雾处理大豆叶片18 h后接种SMV,与对照相比显著降低了SMV病情指数。利用Quantita-tive RT-PCR检测低聚木糖处理的大豆叶片中的防卫相关基因的表达情况,与对照相比,防卫基因PR2、PR10、PR12和LOX2在低聚木糖处理12 h时有最高的相对表达量;PR3在低聚木糖处理18 h时有最高的相对表达量;PR1、PAL、PPO和CHS在低聚木糖处理24 h时显著上调表达。研究表明,低聚木糖激活了大豆SA信号通路和JA信号通路,从而提高了大豆对SMV的抗性。
Treated soybean with different concentrations of xylo-oligosaccharide to study the effect of xylo-oligosaccharide on growth and resistance to soybean mosaic virus(SMV)in soybean.The results of greenhouse experiment showed that 18 days after the seed of soybean were dealt with the xylo-oligosaccharide solution,compared with the contol,50 mg·L-1 xylo-oligosaccharide obviously increased the height,fresh weight,and main root length of soybean seedlings.Inoculated with SMV after 18 hours of pre-atomized the soybean leaves with xylo-oligosaccharide,compared with the contol,the disease index of soybean mosaic virus was obviously reduced.Using Quantitative RT-PCR to test the expression of the defense-related gene of the soybean leaves dealt with xylo-oligosaccharide,compared with the contol,the defense-related gene PR2,PR10,PR12 and LOX2 had the highest related expression quantity at the time of 12 hours,PR3 at 18 hours,but PR1,PAL,PPO,and CHS at 24 hours.Results suggest that xylo-oligosaccharide can active salicylic acid(SA)and jasmonic acid(JA)signaling pathways of soybean,and enhances its resistance against SMV.
