雪芙蓉胶囊对大负荷训练大鼠运动能力及大脑伏隔核c-fos表达的影响

Effects of Xuefurong Capsule on the Sports Ability and c-fos Expression within Nac in Overload Training Rats

目的:观察中药雪芙蓉胶囊对雄性大鼠力竭跑台时间、血清雄激素及乳酸水平、血浆pH值及大脑伏隔核c-fos表达的影响。方法:将30只雄性成年大鼠随机分为3组,即安静对照组、训练组、中药训练组。中药训练组每天灌服雪芙蓉胶囊(5 mg/kg),8周后测试大鼠力竭跑台时间,检测血清睾酮、乳酸含量及血浆pH值,用免疫组织化学方法结合图像半定量方法对c-fos神经元的数量、面积及灰度进行测量和分析。结果:与训练组比较,中药训练组大鼠力竭跑台时间延长、血清睾酮及血浆pH增高(P<0.01,P<0.05,P<0.05)、血清乳酸下降(P<0.05);伏隔核c-fos免疫阳性细胞数量和阳性产物面积表达均降低(P<0.05),灰度没有显著差异(P>0.05)。结论:中药雪芙蓉胶囊可提高大鼠的运动能力,延缓中枢疲劳,机理可能与此中药提高力竭运动大鼠血清睾酮水平及血浆pH值,同时降低大鼠血清乳酸水平及大脑伏隔核c-fos的过度表达有关。

Objective： To explore the effects of Xuefurong capsule on the time of exhausted treadmill, the level of serum testosterone,lactic acid （LA）, the PH of blood plasma, and expression of c-fos within nucleus aeeumbens （Nac） Of brain in the male rats. Method： 30 SD rats were randomly divided into 3 groups： the sedentary control group,the training group,and the Chinese-medicine and training group. The Chinese-medicine and training group were forced to take Xuefurong capsule every day （5mg/kg） and lasted 8 weeks. Then the time of exhausted treadmill of rats was tested, the level of serum testosterone, the LA and the PH of blood plasma were examined, c-los expression within Nac was detected and analysised by using ABC immunohistochemical technique and semi quantitative. Result： Compared with the training group, the time of exhausted treadmill was prolonged（P 〈 0.01 ）, the level of testosterone and PH of blood plasma increased （ P 〈 0.05, P 〈 0.05）, the level of LA de- creased（P 〈 0.05） ; The amount and area of c-fos within Nac obviously decreased（ P 〈 0.05）, the gray degree was not significant different（ P 〉 0.05） in the Chinese medicine and training group. Conclusion： Taking Xue- furong capsules may enhance the sports ability, its mechanism might be that Xuefurong capsule increase the level of serum testosterone and PH blood of plasma, decrease the level of serum LA and excessive expression of c-fos within Nac at the same time.
