丝裂霉素C对瘢痕疙瘩成纤维细胞增殖的影响

摘要 目的探讨丝裂霉素C(MMC)对瘢痕疙瘩成纤维细胞(KFB)增殖的影响。方法采用不同浓度的MMC作用于体外培养的FB,采用四唑盐比色(MTT)方法,观察MMC对体外培养的KFB增殖活性的影响。采用RT-PCR方法,检测MMC对体外培养的KFB TGF-β1 mRNA表达的影响。结果 MTT显示在24 4、8、72 h时间段内,随着培养时间的增加,细胞生长抑制率呈现上升趋势,作用48 h、72 hKFB与24 h相比KFB有极显著差异(P<0.01),作用72 h与48 h相比,有上升趋势,但差异比不显著,24 h为最佳作用时间。RT-PCR方法显示:MMC干预组TGF-β1 mRNA相对表达量明显低于正常对照组,从12.5μg/mlMMC开始具有显著的统计学差异(P<0.01),随MMC浓度增大,MMC干预组TGF-β1 mRNA相对表达量呈逐渐降低趋势。结论 2.5~200μg/ml范围的MMC可有效抑制体外培养的病理性KFB的增殖活性,当浓度为200μg/ml时作用尤为明显,MMC对TGF-β1 mRNA的表达具有明显减弱效应。 Objective To study the effect of mitomycin C (MMC) on cell proliferation of keloid fibroblasts (KFBs). Methods The different concentration of MMC was used to KFB, the activity of KFB and TGF-β1 mRNA expression were tested. Results At 24, 48, 72 h, the growth inhibition rate was increased, TGF-β1 mRNA expression was gradually decreased with the MMC concentration increased. Conclusions MMC could attenuate the expression of TGF-β1 mRNA.
