摘要
目的从药效药理学角度评价主动口服给药及灌胃给药的异同,为今后两种给药方法的选择提供实验依据。方法将氪激光诱导发生脉络膜新生血管的C57BL/6小鼠随机分为4组:灌胃治疗组,灌水对照组,饮药治疗组,饮水对照组,以正常小鼠作正常对照,不做任何处理。观察小鼠的一般情况,分别在治疗后第7天,第14天,第28天,采用ELISA检测外周血中的血管内皮生长因子(vascular endothelial growth factor,VEGF)含量,并在治疗后28 d用FFA检查观察脉络膜新生血管(choroidal neovascularization,CNV)面积。结果治疗过程中灌胃给药小鼠较主动口服给药小鼠体重偏低,精神及活动度稍差,毛色也较差;给药后第7天,14天,灌胃治疗组及饮药治疗组外周血的VEGF含量均低于相应的对照组及正常组(P<0.05),灌胃治疗组和饮药治疗组之间比较无明显差异(P=0.179);治疗后第28天行小鼠FFA检查,灌胃治疗组造影后期CNV渗漏面积明显小于灌胃对照组(P=0.033),饮药治疗组也明显小于饮水对照组(P=0.016),但2个治疗组之间无明显差异(P=0.701)。结论主动口服给药可以达到与灌胃给药相似的治疗结果,对体积小的动物以及需要长期给药的动物实验,主动口服给药也是一种有效的给药方法。
OBJECTIVE To compare and evaluate the pharmacodynamic action of voluntary oral administration with intragastric gavage administration.METHODS C57BL/6 mice were induced CNV(choroidal neovascularization)by krypton laser and randomly divided into 4 groups: intragastric gavage control group,intragastric gavage treatment group,drinking water control group,drinking decoction treatment group.Ten normal mice were used as normal control without any treatment.The general state of health was observed.The VEGF(vascular endothelial growth factor)levels in peripheral blood were determined respectively by ELISA on day 7,14 and 28 after treatment,and the CNV leakage area was examined with FFA on day 28 of the treatment.RESULTS The weights of mice in intragastric gavage groups were lower than that of the voluntary oral administration groups.And the spirit condition,the activity as well as the color pattern of the mice in intragastric gavage groups were also worse than that of the voluntary oral administration groups.The VEGF level in peripheral blood of the two treatment groups were significantly lower than that of its corresponding control groups and normal group on day 7 and day 14 after treatment(P0.05).There were no significant differences between intragastric gavage groups and voluntary oral administration groups(P=0.179).The CNV leakage area of FFA in the two treatment groups were significantly smaller than that of its corresponding control groups on day 28 after treatment(P0.05).There was no significant difference between the intragastric gavage group and the voluntary oral administration group(P=0.701).CONCLUSIONS Voluntary oral administration could have the same pharmacodynamic action as the method of intragastric gavage.For the long-term treating animal experiment or the experiment using tiny animals,voluntary oral administration method is an effective alternative.
