电针对局灶性脑缺血再灌注大鼠迟发性细胞凋亡的影响 被引量:4

Effects of Electroacupuncture Against Late-occurred Neuron Apoptosis of Rats with Focal Cerebral Ischemia/Reperfusion

摘要 目的观察电针对局灶性脑缺血再灌注大鼠缺血区神经细胞凋亡的保护作用及机制。方法雄性SD大鼠随机分为假手术组、模型组、电针组,采用线栓法栓塞大脑中动脉60 min后拔出线栓恢复血流,电针组在缺血再灌注2h后开始电针刺激百会、水沟、足三里穴30 min,每隔12 h重复刺激1次,共7次。再灌注72 h后,TUNEL法检测缺血区神经细胞凋亡,免疫印迹法检测缺血区神经细胞凋亡蛋白Bax、Bcl-2的表达。结果电针组大鼠脑组织神经细胞凋亡数目较模型组明显减少(P<0.01),促凋亡蛋白Bax表达降低(P<0.05)、抗凋亡蛋白Bcl-2表达增高(P<0.01)。结论电针刺激可明显减少脑缺血再灌注大鼠脑组织神经细胞凋亡数量,通过调节Bax/Bcl-2的比值来实现对缺血性神经元的保护作用。 Objective To observe the effects of electroacupuncture (EA) against apoptosis in rat with focal cerebral ischemia/reperfu- sion (IR) injury and its mechanism. Methods Male SI) rats were randomy divided into sham operation group, model group and EA group, they were modified the middle cerebral artery occlusion(MCAO) models with nylon monofilament, the filament was left in place for 60 min and then withdrawn to let the blood reperfuse. EA group was stimulated with EA at the acupoints of Baihui, Shuigou, Znsanli for 30 min 7 time, repeated the stimulation every other 12 h. After 72 h of reperfusion, TUNEL staining was performed to observe the effect of EA against neuron apoptosis after IR, Western blotting was performed to observe the effect of EA against the apoptins of Bax and Bcl -2. Results Neu- ron apoptosis in brain tissue of the EA group significantly alleviated than the model group. The ratio of Bax/Bcl - 2 was significantly reduced than the model group. Conclusion The stimulation of EA can significantly reduce the number of apoptotie neurons in brain tissue, and the protective effect is realized by adjusting the rate of Bax and Bcl -2.
