摘要
目的研究PTEN在三氧化二砷(arsenictrioxide,As2O3)诱导肝癌细胞凋亡中的作用。方法采用四甲基偶氮唑盐(MTT)方法检测As2O3对SMMC-7721细胞增殖的抑制作用;流式细胞仪定量检测各组细胞的凋亡率;Westernblot方法检测各组细胞中PTEN蛋白的表达情况;用PTENsiRNA转染SMMC-7721细胞,观察其对As2O3诱导细胞凋亡作用的影响。结果As2O3能显著抑制SMMC-7721细胞存活率,其抑制率呈剂量依赖关系;As2O3剂量依赖性诱导肝癌细胞凋亡;Westernblot检测显示As2O3明显增加细胞中PTEN蛋白的表达;PTENsiRNA转染可减弱As2O3对SMMC-7721细胞凋亡的诱导作用。结论As2O3诱导SMMC-7721细胞凋亡可能与其诱导PTEN表达有关。
Objective To investigate the effect of PTEN on the apoptosis of hepatoma carcinoma cell line induced by arsenic trioxide ( As2O3 ). Methods The effects of As2O3 on the cell proliferation was measured by methyl thiazolyl tetrazolium(MTT) , apoptosis was determined by flow eytometry, and the expression of P'FEN protein was measured by western blot,and the effect of PTEN inhibition by siRNA on apoptosis of As2O3 -treated SMMC -7721 cells was also examined. Results As2O3 inhibited the proliferation of SMMC -7721 cells in a dose dependent manner. As2O3 also induced apoptosis in a dose dependent manner. As2O3 significantly increased PTEN expression. Transfecting cells with PTEN siRNA significandy attenuated the induced apoptoSis by As2O3. Conclusion It is suggested that As2O3 could induce the apoptosis of SMMC -7721 cells via upregulating PTEN.
