油茶叶中黄酮类物质的提取及纯化工艺研究 被引量:12

摘要 采用超声波萃取法,研究油茶叶黄酮类化合物的提取工艺,从而为油茶树叶工业化提取黄酮类功能性成分提供科学依据。通过单因素和正交试验确定了超声波功率、乙醇体积分数及提取时间、料液比的最佳参数。结果表明,超声波功率450W、乙醇体积分数60%、提取时间为20min,料液比为1∶20提取效果最佳。在此条件下,老叶中的黄酮类化合物含量>中叶中的黄酮类化合物含量>嫩叶中的黄酮类化合物含量。选用AB-8大孔树脂对油茶叶黄酮提取液进行分离纯化。选择上样速度为3BV/h,上液浓度为2.0mg/mL进行吸附,选择3BV90%的乙醇浓度和1.5BV/h的洗脱流速进行解吸试验。提取液经纯化后,黄酮类含量从13.1%提高到27.2%,提高了2.08倍。 The extraction of flavones from Camellia oleifera abel leaves by ultrasonic wave was studied. The results pro- vided scientific basis for industrial production of the functional flavonoids from Camellia oleifera abel leaves. By single factor and orthogonal experiments, the optimum conditions of ultrasonic wave power, alcohol concentration, extraction time and the ratio of solid and liquid were determined. The optimal conditions were: 60% alcohol, ultrasonic wave power at 450W, extraction time 30min and the ratio of solid and liquid 1 : 20. Under the above conditions, the con- tent of flavonoids in the old leaf 〉 the content of flavonoids in the middle leaf 〉 the content of flavonoids in the young leaf. AB -8 resin was used to separate and purify the total flavones. Sample flow rate was 3BV/h, the solution con- centration was 2. 0mg/mL for absorption, 3BV 90% ethanol and 1.5BV/h of elution was the flow rate for desorption. The purity of flavonoids was increased from 13.1% to 27.2%.
