摘要
目的探讨高脂膳食对小鼠以及槲皮素对高脂膳食小鼠学习记忆能力的影响。方法 60只昆明种雄性小鼠,随机分为对照组(正常膳食),高脂膳食组(含18%猪油),槲皮素低剂量组(高脂膳食+0.005%槲皮素)和槲皮素高剂量组(高脂膳食+0.01%槲皮素)。12w后进行Morris水迷宫测试小鼠的学习记忆能力,检测小鼠全血和脑组织的自由基和氧化应激指标并用荧光定量PCR检测海马脑源性神经生长因子(BDNF)的相对表达量。结果高脂膳食使小鼠的学习记忆能力显著下降(P<0.05),脑组织抗氧化能力显著降低(P<0.01),血浆和脑组织丙二醛(MDA)、自由基水平显著升高(P<0.05),海马BDNF的相对表达显著降低(P<0.05)。添加槲皮素后脑部的氧化还原状况得到显著改善(P<0.05)槲皮素高剂量组BDNF基因的相对表达量显著升高(P<0.05)。结论高脂膳食可导致慢性氧化应激,损伤小鼠的学习记忆能力。槲皮素可改善脑部氧化还原状态,增加BDNF的表达,从而改善小鼠的学习和记忆能力。[营养学报,2012,34(4):353-357]
Objective To explore the effect of quercetin on learning and memory of mice administered high fat diet.Methods Sixty KM mice were randomly divided into four groups and fed ordinary diet(CD),high-fat diet(HFD),high-fat diet with 0.005% quercetin(LQD) or 0.01% quercetin(HQD) respectively for 12 w.Performance of spatial learning and memory was measured by Morris water maze and redox status index of plasma and brain were evaluated.The expression of brain-derived neurotrophic factor(BDNF) in hippocampus was determined by real time PCR.Results HFD significantly decreased the performance of learning and memory(P0.05),increased oxidative stress and reduced the relative expression of hippocampus BDNF in mice(P0.05).Quercetin improved the learning and memory of HFD mice.Conclusion HFD may induce chronic oxidative stress,and impair spatial learning and memory in mice.Quercetin can prevent the oxidative damage and normalize the expression of BDNF and improve the learning and memory.
