摘要
目的探讨溴甲酚绿法测定血清白蛋白标准物与反应时间对测定结果的影响,以选择最佳标准物与反应时间。方法用全自动生化分析仪,分别以白蛋白标准液和定值参考血清为标准物,测定其反应时间30、60、120、180、300s血清中白蛋白含量,并作对比分析。结果以白蛋白标准液和定值参考血清作标准物,其测定值两者比较差异有统计学意义(P<0.05);以白蛋白标准液为标准物,其反应时间60、120、180、300s与30s组测定值比较差异有统计学意义(P<0.05或P<0.01);而以定值参考血清作标准物,其测定反应时间30、60、120s三者之间结果比较差异无统计学意义(P>0.05),180、300s与30s组测定值比较差异有统计学意义(P<0.05或P<0.01)。结论溴甲酚绿法测定血清白蛋白,应以定值参考血清作标准物,反应时间可选30、60s或120s。
Objective To explore the effects of standard materials and reaction time on the determination re- sul in the bromocresol green method for determining serum albumin,in order to select the best standard material and reaction time. Methods Automatic biochemical analyzer was used, and the albumin standard solution and the eon- stant reference serum were used as a standard reference materials. The measured response time were :30 seconds, 60 seconds, 120 seconds, 180 seconds ,300 seconds. Tile measured serum albumin contents were compared and analyzed. Results When albumin solution and the standard reference serum as the standard materials, the measured results be- tween the two materials had significant differences (P 〈 0.05 ). When albumin standard solution as the standard refer- ence material. The measured results had significant difference between 30 seconds and other time (P 〈 0.05 or 〈 0. 01 ), and when constant reference serum as the standard reference material, the measured results between 30 sec- onds and 60 seconds, 120 seconds showed no significant diffierence ( P 〉 0. 05 ), but significant difference between 180 seconds, 300 seconds and 30 seconds group (P 〈 0.05 or 〈 0.01 ). Conclusion In determination of serum albumin by bromocresol green method, constant reference serum should be used as the standard material, and 30 seconds,60 seconds or 120 seconds should be selected for the optional reaction time.
