摘要
目的:观察知柏地黄汤合桃红四物汤对CsA肾毒性模型大鼠的肾保护作用。方法:予30mg/kg剂量的CsA灌胃制作大鼠CsA肾毒性模型,同时分别予10mg/kg贝那普利、2g/mL中药药液大鼠灌胃以预防肾毒性。予4周末分别观察各组大鼠肾功能、肾组织病理变化,免疫组化观察各组大鼠肾组织TGFβ1的表达变化。结果:与模型组相比,中药组大鼠血肌酐降低(28.3±3.8 VS 33.1±5.2,P=0.052),病理显示中药组大鼠肾组织空泡变性较模型组改善[变性范围(8.2±2.2)%VS(15.6±3.7)%,P<0.05],炎症细胞浸润及间质纤维化亦减轻;免疫组化显示,中药组大鼠肾组织TGFβ1的表达降低。结论:知柏地黄汤合桃红四物汤具有保护和改善CsA慢性肾毒性大鼠肾功能的作用。其机制有可能与降低肾组织TGFβ1的表达有关。
Objective:To investigate the therapeutic effect of Zhibai Dihuang Decoction combined with Taohong Siwu Decoction on nephrotoxicity of cyclosporin A(CsA).Methods:Rats were on low-salt diet and CsA was administered by gastric gavage at a dose of 30mg/kg once a day for 28 days.Zhibai Dihuang Decoction combined with Taohong Siwu Decoction or benazepril was given to these rats in an appropriate dose.The blood and urine sample were collected at 4 weeks and renal function and total urine protein were examined.Renal morphology was observed and immunohistochemical examination was performed to detect the expression of TGFβ1 in renal tissues.Results:Compared with model group,serum creatinine level was attenuated with the use of tranditional Chinese medcine(28.3±3.8 VS 33.1±5.2,P=0.052).On histopathology tranditional Chinese medcine attenuated tubular cells vacuolations[(8.2±2.2)% VS(15.6±3.7)%,P0.05]as well as interstitial damage including interstitial inflammation and interstitial fibrosis.The expression of TGFβ1 was also reduced in renal tissues with the use of tranditional Chinese medcine.Conclusion:Zhibai Dihuang Decoction combined with Taohong Siwu Decoction can attenuate the structural and functional changes of kidney in CsA nephrotoxicity rats.The mechanism may be related to reducing renal TGFβ1 expression.
