VNP减弱NE刺激的心肌细胞c-fos表达 被引量:3

摘要 目的 :研究血管利钠肽 (VNP)对去甲肾上腺素 (NE)刺激的心肌细胞c fos表达的影响。方法 :分离纯化SD乳鼠心肌细胞 ,随机分为四组 :对照组、NE组、VNP组和VNP +NE组。以免疫组织化学染色方法观察各组c fos的表达情况 ,采用放射免疫方法测定了细胞内cGMP水平。结果 :NE( 10 -6mol/L)可以显著升高Fos样免疫阳性细胞数及染色强度 (P <0 .0 5 ,vs对照组 ) ,VNP( 10 -7mol/L)预处理心肌细胞可以减弱NE的作用 (P <0 .0 5 ,vsNE组 ) ,但fos的表达量仍高于基础水平 (P <0 .0 5 ,vs对照组 )。对照组和NE组细胞内cGMP水平无显著差异 ,VNP组cGMP组水平显著高于对照组和NE组 (P <0 .0 5 )。结论 :VNP能减弱NE诱导的c fos表达 ,其 10 E Aim:This study tested the hypothesis that vasonatrin peptide (VNP)attenuates the expression of cfos in norepinephrine(NE)stimulated cardiac myocytes cultured from neonatal rats.Methods:The cultured cardiac myocytes were divided into four groups: control group,NE group,VNP group,and VNP+NE group.The expression of cfos was measured by the means of immunochistochemistry stain. Furthermore,the effect of VNP on the intracellular level of cGMP was measured by the means of radioimmunoassay. Results:We found that NE(10 -6 mol/L) significatly increased the expression of cfos in cardiac myocytes (P<0.05,vs control group),but VNP(10 -7 mol/L) decreased it,VNP(10 -7 mol/L)increased the intracellular level of cGMP(P<0.05,vs control and NE group).Conclusion:The results reveal that VNP can attenuate the NEinduced expression of cfos in cardiac myocyte which is associated with the changes of cGMP level.
