摘要
目的:探讨甘草苷(Liquiritin,LQ)对膀胱癌细胞T24 p21、PTEN的表达影响及其可能的抗癌机制。方法:实验分组:实验分为对照组,LQ 20μg/mL,50μg/mL,100μg/mL组。用RT-PCR和RT-qPCR检测p21、PTEN的mRNA表达,Westernblot检测LQ对T24细胞内p21、PTEN蛋白的表达影响。结果:与对照组比较,LQ 20μg/mL,50μg/mL,100μg/mL组PTENmRNA和蛋白表达均升高,LQ50μg/mL,100μg/mL组p21mRNA表达升高,LQ 20μg/mL,50μg/mL,100μg/mL组p21蛋白表达明显升高,差异有统计学意义。结论:LQ能诱导T24细胞p21、PTEN的表达,其抗癌机制可能与此相关。
Objective: The present study focus on the effects of Liquiritin(LQ) on p21,PTEN expression of human bladder cancer cell line T24 and its potential antitumor mechanism.Methods: Four groups including control,LQ20μg/mL,LQ50μg/mL,and LQ100μg/mL group were set up in this study.p21 and PTEN mRNA expression of T24 cells was detected by using RT-PCR and RT-qPCR.Expression of p21 and PTEN protein was analyzed by western blot.Results: Compared with the control,after treatment of LQ of 20μg/mL,50μg/mL,100μg/mL,expression of p21 mRNA and protein both increased,and PTEN mRNA and protein also increased.Conclusions: The above results indicated that: LQ induced p21 and PTEN expression,and this maybe related antitumor effect of LQ.
