层粘连蛋白、纤粘连蛋白及Ⅳ型胶原抗血清对小鼠胚泡着床的影响 被引量:2

摘要 文中报道了子宫角单次注射层粘连蛋白 (LN)抗血清、纤粘连蛋白 (FN)抗血清以及Ⅳ型胶原 (ColⅣ )抗血清对小鼠胚胎植入的作用。选用 6 0只 30 g左右的昆明品系雌性成年小鼠 ,与有生殖能力的雄性小鼠按 1∶1合笼 ,翌晨检查外阴 ,以出现阴栓为妊娠第 1天。于妊娠第 3天下午 ,给每个雌性小鼠左侧子宫角单次注射 5 μlLN抗血清、FN抗血清或ColⅣ抗血清作为实验组 ;给同一小鼠右侧子宫角注射 5 μl正常兔血清 (Normalrabbitserum ,NRS)作为对照组。于妊娠第 8天剖检受试雌鼠 ,观察并统计每侧子宫角着床的胚胎数 ,并以t检验。实验发现 :LN抗血清和正常兔血清处理的子宫角 (n =19)着床胚胎数 (平均值±标准差 )分别为 2 0 7± 1 0 6和 5 15± 1 14;FN抗血清和正常兔血清处理的子宫角 (n =2 4)着床胚胎数分别为 1 83± 1 46和 5 6 6± 2 0 2 ;ColⅣ抗血清和正常兔血清处理的子宫角 (n =17)着床胚胎数分别为 2 0 7± 1 0 3和 5 71± 2 0 2。三组实验侧着床胚胎数均极显著低于对照侧的着床数 (P <0 0 1)。结果表明 ,子宫角单次注射 5 μlLN抗血清。 Effects of single injection of antiserum against laminin (LN), fibronectin (FN) or type Ⅳ collagen (Col Ⅳ) into uterine horns on embryo implantation in mouse were studied. More than sixty adult female mice ( around 30 g) of Kunming white strain were maintained at 22~25 ℃ on a 14 h light~10 h dark cycle. Males and females were placed 1∶1 overnight to impregnate. Mating was evidenced by the appearance of a vaginal plug on the following morning. Plugged females designated day 1 of pregnancy were caged separately. In the afternoon of day 3 of pregnancy female mice were injected with 5 μl of the antiserum into one horn (experiment group) of uteri and with 5 μl normal rabbit serum (NRS) into another horn (control) in the same mouse. The numbers of embryos implanted were counted and recorded on day 8 of pregnancy to study the effects of the antisera on embryo implantation in vivo . The data were analysed by student's t test. The numbers of embryos implanted in the horns treated with antisera against LN (2 07±1 06, n=19), against FN(1 83±1 46, n=24) and against Col Ⅳ(2 07±1 02, n=17) were significantly less than ( P <0 01) those in corresponding controls (5 15±1 14, 5 66±2 08 and 5 71±2 02). It may be concluded from our experiments that single injection of 5μl antiserum against LN, FN or Col Ⅳ into uterine horns may inhibit embryo implantation in mouse.
