摘要 目的:建立枸杞子中甜菜碱含量的HPLC-ELSD测定方法。方法:采用Hypersil NH2色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-四氢呋喃-0.2%三氟乙酸(30:60:10),流速为1.0 mL.min-1,柱温为25℃。结果:甜菜碱进样量为1.28~12.8μg时,其与色谱峰面积具有良好的线性关系(r=0.999 2);平均加样回收率为98.60%,RSD为2.8%(n=6);测得样品中甜菜碱含量为0.528 1~1.205 4 mg.g-1。结论:该HPLC-ELSD法简便,准确度高,可用于枸杞子的质量控制。 Objective:To establish a HPLC-ELSD method for the determination of the content of betaine in Lycii Fructus.Methods:The analysis was performed on Hypersil NH2 column(250 mm×4.6 mm,5 μm).The mobile phase was composed of methanol,tetrahydrofuran and 0.2% trifluoroacetic acid with the volume ratio of 30 ∶60 ∶10.The flow rate of mobile phase was 1.0 mL ·min-1 and the column tempe-rature was and 25 ℃.Results:The calibration curve showed a good linearity with the range of 1.28~12.8 μg(r=0.999 2) for betaine.The average recovery was 98.60%,with the RSD of 2.8%(n=6).The content of betaine is determined as 0.528 1~1.205 4 mg ·g-1.Conclusion:The method is simple and accurate.It can be used to control the quality of Lycii Fructus.
