雌激素对高糖诱导的HepG2细胞ACAT2表达的影响

摘要 目的探讨雌激素对高糖诱导的酰基辅酶A:胆固醇酰基转移酶2(ACAT2)在HepG2细胞中表达的影响。方法将培养好的HepG2细胞以0.7×106/L接种于六孔培养板内,加入无血清培养基继续培养24 h,将细胞分为四组,对照组加入正常培养基,甘露醇组计入终浓度为25 mmol/L甘露醇溶液,高糖组加入的葡萄糖溶液培养基孵育12 h;雌激素组用含总浓度25 mmol/L葡萄糖的培养基与细胞孵育4 h后,再加入终浓度为1×10-8mol/L的17β-雌二醇孵育8 h。RT-PCR和Western blot法检测各组ACAT2的表达水平。结果高糖组HepG2细胞ACAT2的表达明显高于对照组和雌激素组(P<0.001),雌激素组与对照组、甘露醇组比较无统计学意义。结论雌激素可下调高糖诱导的ACAT2表达。
