樟芝胶囊生产工艺中发酵液与醇沉多糖中腺苷含量的测定 被引量:2

摘要 利用高效液相色谱法建立了樟芝胶囊生产工艺中发酵液与醇沉多糖中腺苷含量的测定方法。采用色谱柱Agilent Eclipse XDB-C18(4.6 mm×150 mm,5μm),磷酸盐缓冲溶液(pH 6.47)-甲醇(90∶10)为流动相,流速1.0 ml/min,检测波长260 nm,柱温为室温。结果表明,腺苷的线性方程为Y=29160X-4972,在1.17~37.5 mg/L范围内线性关系良好,r=0.9999。平均回收率为102.8%,RSD=1.29%。本方法简单、快速、准确,可作为樟芝胶囊生产过程中腺苷含量测定的一种有效方法。 An HPLC method was established to detect the adenosine content in fermentation liquid and ethanol sediments of polysaccharide during the production process of Antrodia camphorata capsules. The reverse - phase bonded Agilent Eclipse XDB - C18 (4.6 mm × 150 mm, 5μm) was selected, and the mobile phase consisted of phosphate acid buffer solution (pH 6.47 ) and methanol at the volume rate of (90: 10) with the flow rate as 1.0 ml/min. The detective wavelength was 260 nm, and the column temperature was kept as the room temperature. The results showed that the linear equation for adenosine detection was Y = 29160X - 4972 with the linear range of 1.17 -37.5 mg/L and r as 0. 9999. The average recovery rate was 102.8% and the RSD was 1.29%. In conclusion, the method was simple, sensitive and accurate, so it could be used to determine the adenosine content in fermentation solution and ethanol sediments during production process of Antrodia camphorate capsules.
