摘要 目的:研究黄芪多糖对COLO205人结肠癌细胞株增殖及凋亡的影响。方法:黄芪多糖(25μg/mL、50μg/mL、100μg/mL)与结肠癌细胞共同培养24、48、72h,利用MTT法检测细胞增殖抑制率、免疫组化法检测细胞增殖核抗原(PCNA)表达情况、TUNEL法检测细胞凋亡情况。结果:50μg/mL以上浓度的黄芪多糖对体外培养的COLO205人结肠癌细胞株生长具有抑制作用(抑制率>30.0%,P<0.01),并呈量效和时效关系;与对照组相比,黄芪多糖处理组(50μg/mL、100μg/mL)PCNA表达降低,细胞凋亡增加(凋亡指数>31.47%,P<0.01)。结论:黄芪多糖使COLO205人结肠癌细胞增殖减慢,凋亡增加,PCNA表达降低,可能是黄芪多糖抗肿瘤机制之一。 [Objective] To investigate the effect of astragalus polysaccharides on COLO205 colon cancer cells proliferation and apoptosis. [Method] COLO205 colon cancer cells were co-cultured with various of astragalus polysaccharides for 24h, 48h, 72h respectively, and the rate of cell proliferation inhibition was detected by MTY assay. The levels of PCNA expression were detected by immunohistochemistry method, and the cell apoptosis were detected by TUNEL kit. [Result] Astragalus polysaccharides could effectively inhibit the COLO205 colon cancer ceils proliferation in dose-dependent and time-dependent manner, reduce the level of PCNA expression, and induce cell apoptosis. [Conclusion] Astragalus polysaccharides substantially inhibited COLO205 colon cancer cells proliferation, induced apoptosis and down regulated PCNA expression.
