摘要
目的研究激肽释放酶相关肽6(KLK6)对体外乳腺癌细胞迁移及侵袭能力的作用,并探讨其在肿瘤组织中高表达的临床意义。方法运用划痕试验及Transwell细胞侵袭试验,评价人重组KLK6对乳腺癌细胞株MDA-MB-231的迁移及侵袭运动的影响,半定量反转录-聚合酶链反应检测激肽释放酶相关肽6在乳腺癌组织的表达水平,统计学方法分析其高表达的临床意义。结果 KLK6明显增强MDA-MB-231细胞的体外迁移及侵袭能力,KLK6与淋巴转移相关。结论 KLK6是一个潜在的预测乳腺癌肿瘤转移的分子诊断标志物及肿瘤治疗的基因靶点。
Objective To examine the effect of kallikrein-related peptidase 6(KLK6)on migration and invasion of breast cancer cell in vitro,and investigate the clinical significance of enhanced expression of KLK6 in primary tumor.Methods Effect of KLK6 on migration and invasion in breast cancer cell line MDA-MB-231 was elucidated by performing wound-healing and Transwell assay,semi-quantitative RT-PCR was used to detect expression of KLK6 in primary tumor,clinical significance of enhanced expression of KLK6 was analyzed by statistical method.Results KLK6 promoted MDA-MB-231 cells migration and invasion in vitro significantly,enhanced expression of KLK6 was found to be related to lymph node metastasis.Conclusion KLK6 is a potential molecular marker to predict tumor metastasis in breast cancer and a feasible gene target for cancer therapy.
