摘要
目的:研究地黄不同炮制品中水苏糖的含量差别以及水苏糖对肿瘤的抑制活性。方法:从地黄不同炮制品中提取水苏糖并计算含量;用MTT法和台盼蓝染色法检测不同浓度水苏糖体外对HepG-2和SGC-7901肿瘤细胞的抑制活性;观察水苏糖与环磷酰胺合用对小鼠H22肝癌细胞体内生长的影响。结果:鲜地黄、生地黄、熟地黄中水苏糖含量依次为26.45%、9.16%、5.98%;MTT法和台盼蓝染色法实验结果均表明水苏糖对HepG-2和SGC-7901肿瘤细胞具有明显的抑制作用(P<0.05),其中水苏糖对HepG-2人肝癌细胞作用比SGC-7901人胃癌细胞作用更明显。环磷酰胺与不同剂量水苏糖合用对H22小鼠均有抑制作用(P<0.05),抑瘤率分别为57.59%、68.06%和76.44%。结论:鲜地黄中水苏糖含量最高,其次是生地黄,熟地黄中含量最低;水苏糖体外对HepG-2和SGC-7901肿瘤细胞具有明显的抑制作用;水苏糖能明显增强环磷酰胺的抑瘤作用。
Objective:To study the content difference of stachyose in different processed products of rehmannia and the stachyose’s inhibitive activity on tumor.Methods:To extract the stachyose from rehmannia’s different processed product and to caculate the content; In vitro, we used MTT assay and trypan blue staining method to observe the inhibitive activity of stachyose which has different concentrations in HepG-2 and SGC-7901 tumour cells;Using stachyose and Cytoxan together in H22 hepatoma carcinoma cells to observe cell’s growth in vivo.Results:In fresh rehmannia, unprocessed rehmannia and prepared rehmannia root, the content of stachyose were 26.45%、9.16%、5.98% successively. MTT assay and trypan blue staining method both revealed that stachyose significantly inhibited the HepG-2 and SGC-7901 tumour cells (P0.05), rehmannia’s depressant effect in HepG-2 human hepatoma carcinoma cells was more obviously then that in SGC-7901 human gastric cancer cells. Cytoxan was combined with different doses of stachyose in H22 mice,which were all have the inhibitive activity, inhibition rates were 57.59%、68.06% and 76.44% successively.Conclusion:The content of stachyose in fresh rehmannia is the hightest, the second is the unprocessed rehmannia, and the prepared rehmannia root has the lowest; In vitro, stachyose has significant inhibitive effect on HepG-2 and SGC-7901 tumor cells; stachyose can enhance the antitumor effect of Cytoxan.
