PCR-DGGE研究亚硝化-电化学生物反硝化全自养脱氮工艺细菌多样性 被引量:2

Bacterial diversity of complete autotrophic nitrite-nitrification and electrochemical bio-denitrification combined process using PCR-DGGE

摘要 采用分子生物学手段PCR-DGGE技术对亚硝化-电化学生物反硝化全自养脱氮工艺细菌的多样性进行了研究。结果表明,亚硝化段内主要的细菌种群为相似于Nitrosomonas sp.(AJ224410)和Nitrosomonas sp.NM41(AF272421)的种群,相似性分别为97%和94%;电化学生物反硝化段细菌类群主要有β-proteobacteria类群、γ-proteobacteria类群和Chlo-roflexi类群。填料上生物膜细菌种群较底部泥水混合物丰富,两者细菌种群相似性为75%;底部泥水混合物样中存在与厌氧氨氧化菌Brocadia anammoxidans(AF375994)相似性为93%的菌种,而填料上生物膜中存在与Thioalkalivibrio sp.K90mix(EU709865)和Thiobacillus thioparus(AJ243144)相似性分别为94%、97%的菌种,其中Thiobacillus thioparus(AJ243144)是典型的硫自养反硝化菌,表明填料上生物膜中有大量的硫自养反硝化菌。 The bacterial diversity in the complete autotrophic nitrite-nitrification and electrochemical bio-denitrification combined process was studied by PCR-DGGE method.The results indicated that the dominant bacteria was similar to the Nitrosomonas sp.(AJ224410)and Nitrosomonas sp.NM41(AF272421)in nitrite-nitrification reactor.The dominant bacterial communities in the electrochemical bio-denitrification reactor were β-proteobacteria,γ-proteobacteria and Chloroflexi bacterium.The bacterial community richness of the electrochemical bio-denitrification reactor bottom sludge was slightly more than that of the packing granule biofilm.The bacterial community similarities of the bottom sludge and granule biofilm was 75%.The anaerobic ammonium oxidation bacterium,which similarity to Brocadia anammoxidans(AF375994)was 93%,was found in the electrochemical bio-denitrification reactor bottom sludge.While the bacteria similarities to Thioalkalivibrio sp.K90mix(EU709865)and Thiobacillus thioparus(AJ243144)were 94% and 97%,respectively in the biofim.Thiobacillus thioparus(AJ243144) was the typical sulfur autotrophic denitrification bacteria,which indicated that a large amount of sulfur autotrophic denitrification bacteria was in the packing granule biofilm.
