桑枝对小鼠脑缺血再灌注损伤的保护作用 被引量:7

摘要 目的:观察桑枝对小鼠脑缺血再灌注损伤的保护作用。方法:将小鼠分为4组,假手术组;模型组(分离两侧颈总动脉及迷走神经,用微型动脉夹同时夹闭双侧颈总动脉,制作小鼠脑缺血再灌注模型);阳性药组给以尼莫地平24mg/kg,受试药组给以桑枝水提醇沉液14g生药/kg,均灌胃给药7天,24h后取材,匀浆,测脑组织Evan's蓝含量及抗氧化酶SOD活性与MDA水平的变化。结果:缺血再灌注后,脑组织Evan's蓝含量升高,给药后降低。脑缺血再灌注组SOD活性降低,MDA含量增高,而治疗组SOD活性增高,MDA水平降低。结论:桑枝能增加脑缺血再灌注小鼠SOD的活性,改善脑血流,减轻血脑屏障的损伤,对脑缺血再灌注损伤具保护作用。 Objective :To investigate the protection of Mulberry on cerebral ischemia -reperfusion injury in mice brain tissue. Methods :The mice were divided into four groups:control group; model group( separate carotid artery and the vagus nerve,use micro- artery occlusion folder on bilateral carotid artery, reproduce mice brain ischemia -reperfusion model), positive drug group (nimodipine 20mg/kg) ; tested drug Mulberry group (20mg/kg). Every group took intragastric adminis- tration,once a day for seven days. Brain tissue Evan~ Blue content and antioxidant enzymes and other changes were detec- ted. Result:Evan~ Blue content in the brain tissue increased in model group, meanwhile, it decreased after nimodipine and Mulberry administration. SOD activity of cerebral ischemia - reperfusion group decreased and MDA levels increased, but in the treatment group the activity of SOD increased, and MDA decreased. Conclusion:Mulberry can increase the activity of SOD,improve cerebral blood flow and reduce the blood - brain barrier damage in cerebral ischemia - reperfusion of mice.
