摘要 目的:准确、快速测定盾叶薯蓣中薯蓣皂苷的含量。方法:以薯蓣皂苷含量为评价指标,采用高效液相色谱法对薯蓣皂苷的检测条件进行了优化;同时,运用3因素3水平的正交实验,对薯蓣皂苷的提取条件进行了研究。结果:优化后的检测条件为:C18色谱柱(250mm×4.6mm,5μm),柱温30℃,乙腈-水(45∶55),进样量20μL,流速1.0mL.min-1,检测波长210nm。最优提取条件为药材称取量2g,用80%乙醇超声提取40min。结论:优化后的提取及检测条件能快速准确测定盾叶薯蓣中薯蓣皂苷的含量。 Objective:To detect the content of dioscin in Rhizoma Dioscoreae Zingiberensis fast and precisely. Meth- ods:The content of dioscin was valued as an evaluation, the detection conditions of dioscin were optimizated by RP - HPLC ; at the same time, the extraction processes were studied by the orthogonal study of three factors and three levels. Results :The optimal detection extraction condtions were as foUows:the Phenoxent Luna C18 (51.Lm ,250mm 4.6mm)was used at 30~C with gradient elution of acetonitrile -water 45 : 55;the sample size was 20uL; the flow rate was 1.0mL · min-1. The detection wavelength was set at 210nm. The optimal extraction process conditions were as follows :2g of sam- ple is added in 50mL 80% alcohol and ultrasonic time is 40min. Conclusion:The optimized extraction and detection condi- tions could detect the content of dioscin fast and precisely .
