摘要
目的探讨RECK和MMP-2基因在骨肉瘤中的表达及其临床意义。方法采用荧光实时定量PCR(RT-PCR)对我院2001年至2006年45例骨肉瘤手术切除标本中RECK mRNA、MMP-2 mRNA进行检测,同时结合患者的性别、年龄、转移、复发等临床病理特征,分析其表达的临床意义。结果 45例骨肉瘤组织中RECK mRNA和MMP-2 mRNA的相对表达量分别为38.15±14.15和65.00±22.86,两者表达呈负相关(r=-0.740,P=0.000)。RECK mRNA和MMP-2 mRNA表达与性别、年龄、肿瘤部位、组织亚型无关(P>0.05),与肿瘤复发、转移及5年生存期显著相关(P<0.05)。结论 RECK mRNA和MMP-2 mRNA表达与骨肉瘤转移、复发及预后相关,可作为预测骨肉瘤转移、复发及预后的分子指标。
Objective To explore the expressions of RECK mRNA and MMP-2 mRNA in osteosarcoma and their clinical significance.Methods RT-PCR was used to determine the expressions of RECK mRNA and MMP-2 mRNA in 45 osteosarcoma tissues from Nanjing General Hospital of Nanjing Military Command.Results The relative quantitative expressions of RECK mRNA and MMP-2 mRNA were 38.15±14.15 and 65.00±22.86 in osteosarcoma tissues.There was a negative relationship between two genes(r=-0.740,P=0.000).The expressions of RECK mRNA and MMP-2 mRNA were correlated to metastasis,recurrence and prognosis(P〉0.05),but they were not related to age,gender,tumor location and subtype(P〉0.05).Conclusion The expressions of RECK mRNA and MMP-2 mRNA are correlated with recurrence,metastasis and prognosis.They can be relevant biomarkes in predicting metastasis,recurrence and prognosis of osteosarcoma patients.
