摘要
以少根紫萍(Landoltia punctata)为原料,以丙酮丁醇梭菌(Clostridium acetobutylicum)CICC 8012为菌种发酵生产燃料丁醇的研究。筛选出高淀粉含量的少根紫萍,HPLC分析其酸水解液表明,单糖组成有葡萄糖、木糖和半乳糖。糊化后的少根紫萍培养基可以直接发酵,47.50g/L的初总糖发酵可生成9.31g/L的丁醇、13.60g/L的丙酮丁醇乙醇,发酵得率为0.35,其中丁醇占总溶剂的比例为68.46%,比玉米发酵的高14.11%。少根紫萍培养基经降黏后发酵,丁醇和总溶剂分别提高到9.61g/L和14.80g/L,比没降黏的提高了3.22%和8.82%。HPLC分析表明发酵残糖含有以木糖和半乳糖为单糖组成的寡糖和少量的葡萄糖。该研究首次开发了一种新型非粮原料少根紫萍发酵生产燃料丁醇,工艺简单,并且还具有目标产物丁醇在总溶剂的比例高的优点。
Butanol produced from Landoltia punctata by Clostridium acetobutylicum CICC 8012 was studied. High starch content duckweed L. punc- tata was screened and the monosaccharide composition of acidic hydrolysate of L. punctata contains glucose, xylose and galactose was obtained by HPLC analysis. It was found that the gelatinized L. punctata culture medium could be fermented directly. It was observed that 9.31g/L butanol and 13.60g/L acetone butanol ethanol (ABE) was produced from 47.50g/L total sugar of gelatinized culture medium of L. punctata and the fermentation yield was 0.35, in which, butanol accounted for 68.46% of total solvent and 14.11% higher than that fermented with corn starch. The was fermented after, Yield of butanol and ABE reached 9.61g/L and 14.80g/L, respectively, when fermented with viscosity reduced gelatinized culture medium of L. punctata, which showed an improvement of 3.22% and 8.82% respectively. Analysis with HPLC indicated that residual sugar mainly contained polysaceharides that composed of xylose and galactose, and a little glucose. This research firstly developed a new non-raw feedstock on biobutanol fermentation technology, which was simple and had advantages of high ratio of target product-biobutanol.
